Amlodipine + losartan + chlorthalidone is a fixed-dose combination (FDC) small molecule therapy developed by Hanmi Pharmaceutical for the treatment of hypertension. This triple-combination drug integrates three distinct antihypertensive mechanisms: amlodipine, a dihydropyridine calcium channel blocker that induces peripheral vasodilation; losartan, an angiotensin II receptor type 1 (AT1R) antagonist that inhibits the renin-angiotensin-aldosterone system; and chlorthalidone, a thiazide-like diuretic that promotes sodium and water excretion. By targeting multiple physiological pathways simultaneously, the combination is designed to provide superior blood pressure reduction and improved response rates compared to dual therapies or monotherapies. Hanmi also developed a low-dose variant (HCP1803, branded as Amoprel or Amosartan PLUS-L) intended for early-stage or first-line treatment of hypertension.
