amlodipine + olmesartan medoxomil

G3041 is a fixed-dose combination (FDC) therapy developed by Dong-A Pharmaceutical for the treatment of hypertension. It consists of amlodipine, a dihydropyridine calcium channel blocker (CCB), and olmesartan medoxomil, an angiotensin II receptor blocker (ARB). Amlodipine acts by inhibiting the transmembrane influx of calcium ions into vascular smooth muscle and cardiac muscle, leading to vasodilation and reduced peripheral resistance. Olmesartan medoxomil is a prodrug that is hydrolyzed to olmesartan, which selectively blocks the binding of angiotensin II to the AT1 receptor in vascular smooth muscle, thereby inhibiting its vasoconstrictive and aldosterone-secreting effects. The combination of these two antihypertensive agents provides synergistic blood pressure lowering effects. G3041 was developed as a generic or incremental innovation version of the reference drug Sevikar and has been evaluated in Phase I clinical trials for bioequivalence and pharmacodynamic comparability.
