**Amlodipine + valsartan** is an oral fixed-dose antihypertensive combination comprising amlodipine, a dihydropyridine calcium-channel blocker, and valsartan, an angiotensin II type 1 receptor blocker. Amlodipine inhibits L-type calcium-channel-mediated calcium influx in vascular smooth muscle, producing arterial vasodilation, while valsartan blocks angiotensin II signaling at the AT1 receptor, reducing vasoconstriction and aldosterone-mediated sodium retention. The combination is marketed as **Exforge** and is approved for treatment of hypertension, including patients inadequately controlled on either component alone and selected patients likely to require combination therapy.
