Amlodipine + valsartan + rosuvastatin is an oral fixed-dose combination of three small-molecule cardiovascular agents for patients with coexisting hypertension and hyperlipidemia. Amlodipine blocks L-type voltage-gated calcium channels to reduce vascular smooth-muscle tone and blood pressure; valsartan antagonizes the angiotensin II type 1 receptor to inhibit angiotensin II-mediated vasoconstriction and aldosterone signaling; and rosuvastatin competitively inhibits HMG-CoA reductase to lower hepatic cholesterol synthesis and circulating low-density lipoprotein cholesterol. The combination was clinically developed by HK inno.N as CJ-30060, including a completed Phase III trial in hypertensive patients with hyperlipidemia.
