Drug intelligence / Profile preview
AMN082 is a selective, orally active, and brain-penetrant small molecule allosteric agonist of metabotropic glutamate receptor 7 (mGluR7). Discovered by Novartis, it binds to an allosteric site within the transmembrane domain of mGluR7, directly activating receptor signaling, inhibiting cAMP accumulation, and stimulating GTPγS binding. In preclinical research, AMN082 has been widely used as a pharmacological tool to investigate the role of mGluR7 in stress-related central nervous system disorders, showing antidepressant-like and anxiolytic-like effects, modulating stress hormone levels (such as corticosterone and ACTH), and regulating synaptic transmission.
