Amobarbital is a barbiturate derivative with sedative-hypnotic properties, primarily used for the induction of sedation during procedures, short-term management of insomnia, and acute management of refractory tonic-clonic seizures. It is classified as an intermediate-acting barbiturate and acts as a central nervous system depressant. Amobarbital exerts its effects mainly by binding to Gamma-aminobutyric acid type A receptor (GABAA receptor) at sites distinct from both GABA and benzodiazepine binding sites, thereby potentiating the inhibitory action of GABA. This results in decreased neuronal excitability through increased chloride channel conductance, leading to drowsiness, sedation, hypnosis, and anticonvulsant effects. Additionally, it can block AMPA-type glutamate receptors and bind to neuronal nicotinic acetylcholine receptors. Amobarbital is metabolized primarily in the liver via microsomal enzymes and excreted mainly as metabolites in urine[1][2][5][8].
