Overview

This is a combination antimalarial therapy consisting of three components: - **Amodiaquine** (a 4-aminoquinoline antimalarial) - **Artesunate** (an artemisinin derivative) - **Artemether-lumefantrine** (a fixed-dose combination of two antimalarials, artemether and lumefantrine). Each component acts on the malaria parasite at different stages or through different mechanisms. Amodiaquine interferes with the parasite's ability to detoxify heme, leading to toxic buildup within the parasite. Artesunate and artemether are fast-acting derivatives of artemisinin that generate free radicals inside infected red blood cells, rapidly reducing parasitemia. Lumefantrine is a longer-acting partner drug that inhibits heme polymerization in the parasite. This triple therapy is designed for use in areas with high levels of resistance or treatment failure to standard regimens, aiming to improve efficacy and reduce the risk of resistance development by combining multiple drugs with distinct mechanisms. The primary indication is for treatment of uncomplicated *Plasmodium falciparum* malaria.
