Overview

A combination antimalarial regimen consisting of amodiaquine, artesunate, sulfadoxine, and pyrimethamine. This specific combination was evaluated by the Papua New Guinea Institute of Medical Research in a Phase 3 clinical trial (NCT00285662) for Intermittent Preventive Treatment in infants (IPTi). The regimen was administered as two separate treatment arms: sulfadoxine-pyrimethamine plus amodiaquine (SP-AQ) and sulfadoxine-pyrimethamine plus artesunate (SP-AS). The components target the Plasmodium parasite through multiple mechanisms: sulfadoxine and pyrimethamine inhibit folate synthesis by targeting dihydropteroate synthase and dihydrofolate reductase, respectively; amodiaquine interferes with the detoxification of heme; and artesunate generates reactive oxygen species that are toxic to the parasite. The trial aimed to reduce the burden of malaria and associated anemia in infants living in endemic regions of Papua New Guinea.
