Amorolfine is a morpholine antifungal drug primarily used as a topical treatment for onychomycosis (fungal infections of the fingernails and toenails). It acts by inhibiting the fungal enzymes Delta 14-reductase and Delta 7,8-isomerase, which are involved in ergosterol biosynthesis. This inhibition leads to depletion of ergosterol and accumulation of ignosterol in the fungal cell membrane, disrupting membrane integrity and function. Amorolfine is available mainly as a 5% nail lacquer applied once or twice weekly. It has broad-spectrum fungicidal activity against dermatophytes, yeasts, dimorphic fungi, and molds but lacks efficacy for systemic mycoses due to poor systemic absorption. The drug is marketed under several brand names including Curanail, Loceryl, Locetar, and Odenil.
