Overview
Amoxicillin + flucloxacillin is a fixed-dose combination (FDC) antibiotic therapy consisting of two semi-synthetic beta-lactam penicillins. Amoxicillin provides broad-spectrum activity against Gram-positive and Gram-negative bacteria but is susceptible to degradation by beta-lactamase enzymes. Flucloxacillin is a narrow-spectrum, beta-lactamase-resistant penicillin that specifically targets staphylococci, including beta-lactamase-producing strains. Together, they act synergistically to treat mixed infections or infections where the pathogen's resistance profile is uncertain. The combination works by binding to penicillin-binding proteins (PBPs), thereby inhibiting the final transpeptidation step of peptidoglycan synthesis in bacterial cell walls, leading to cell lysis. It is widely used for respiratory tract infections, skin and soft tissue infections, and surgical prophylaxis, particularly in regions where beta-lactamase-producing *Staphylococcus aureus* is prevalent.
