amoxicillin + metronidazole + diclofenac sodium is a multi-drug regimen used as the standard-of-care control in clinical research conducted by the University of Science and Technology, Yemen. This combination is specifically employed to manage postoperative complications, such as pain, trismus (lockjaw), and facial edema, following the surgical extraction of impacted mandibular third molars. The regimen consists of two antibiotics: amoxicillin, a beta-lactam that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), and metronidazole, a nitroimidazole that targets anaerobic bacteria by causing DNA strand breakage. These are paired with diclofenac sodium, a non-steroidal anti-inflammatory drug (NSAID) that provides analgesic and anti-inflammatory effects by inhibiting the enzymes cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2), thereby reducing the synthesis of pro-inflammatory prostaglandins.
