Overview

AMP-220 is a synthetic small molecule analog of illudin S, a cytotoxic sesquiterpene naturally produced by the mushroom *Omphalotus illudens*. Developed by AmpliMed Corporation, AMP-220 was designed as a next-generation DNA-reactive agent with an improved therapeutic index compared to its parent compound. The drug's mechanism of action involves intracellular activation, likely by enzymes such as prostaglandin reductase 1 (PTGR1), to form highly reactive intermediates that create covalent DNA adducts. These adducts interfere with DNA replication and transcription, leading to cell cycle arrest and apoptosis, particularly in tumor cells with deficient transcription-coupled nucleotide excision repair (TC-NER) pathways. AMP-220 reached Phase 1 clinical trials for the treatment of advanced solid tumors, where it demonstrated dose-limiting myelosuppression, but development was ultimately discontinued following the dissolution of its developer.
