Overview

AMP-224 is a recombinant fusion protein that consists of the extracellular domain of human programmed death ligand 2 (PD-L2) fused to an immunoglobulin Fc domain. It acts as an anti-PD-1 agent by specifically binding to the programmed cell death protein 1 (PD-1) receptor on chronically stimulated or exhausted T cells but not on normal activated T cells. By binding to PD-1^high^ T cells and reducing their proliferation, it aims to restore immune function and promote cytotoxic T cell-mediated responses against tumor cells. Unlike traditional PD-1 blocking antibodies that inhibit ligand-receptor interaction directly, AMP-224 depletes exhausted effector T cells expressing high levels of PD-1 and may allow replenishment with functional T-cells[1][8][10]. The drug was developed for use in cancer immunotherapy—primarily metastatic colorectal cancer and other solid tumors—but has not demonstrated significant clinical benefit in trials so far[6][8].
