Overview

AMP-224 + cyclophosphamide is an investigational combination therapy consisting of AMP-224, a fusion protein that targets the programmed cell death 1 (PD-1) receptor, and cyclophosphamide, an alkylating agent used in cancer treatment. AMP-224 acts as a PD-1 pathway modulator by binding to PD-1 on T cells and potentially altering immune responses against tumors. Cyclophosphamide is included at low doses for its immunomodulatory effects and cytotoxic activity against cancer cells. This combination has been studied primarily in patients with metastatic colorectal cancer (mCRC) refractory to standard chemotherapy regimens. In clinical studies, the regimen was generally well tolerated but did not demonstrate significant clinical benefit in terms of objective response rates or survival extension[5][1]. The combination has also been evaluated alongside stereotactic body radiation therapy (SBRT)[5][1].
