Amphotericin B lipid complex is a broad-spectrum antifungal drug used to treat serious, life-threatening fungal infections in patients who are unresponsive to or unable to tolerate conventional amphotericin B therapy[1][2][4]. It consists of amphotericin B complexed with two phospholipids—L-α-dimyristoylphosphatidylcholine (DMPC) and L-α-dimyristoylphosphatidylglycerol (DMPG)—in a 7:3 molar ratio, forming ribbon-like structures[5][6]. The lipid formulation alters the pharmacokinetics and tissue distribution of the drug, resulting in reduced nephrotoxicity compared to conventional formulations. Amphotericin B exerts its antifungal effect by binding to ergosterol in fungal cell membranes, creating pores that lead to cell death.
