**Amplivant-HPV16-SLP**, also called **Hespecta**, is an investigational intradermal therapeutic cancer vaccine developed from two synthetic long peptides derived from immunodominant regions of the human papillomavirus type 16 E6 oncoprotein. Each peptide is covalently conjugated to Amplivant, a molecularly optimized Toll-like receptor 1/2 ligand adjuvant. The conjugate is intended to improve dendritic-cell antigen presentation and induce HPV16 E6-specific CD4-positive and CD8-positive T-cell immunity against HPV16-positive premalignant lesions and cancers, including HPV16-positive oropharyngeal squamous cell carcinoma. In a Phase 1 dose-escalation study, three intradermal vaccinations produced dose-dependent HPV16-specific T-cell responses with predominantly mild injection-site inflammation and transient flu-like adverse events.
