Overview

**Amprenavir + nucleoside reverse transcriptase inhibitors (NRTIs)** is a combination antiretroviral regimen used primarily for the treatment of HIV-1 infection. Amprenavir is a synthetic small molecule that selectively inhibits the HIV-1 protease enzyme, preventing the cleavage of gag and gag-pol polyprotein precursors, thereby producing immature, non-infectious viral particles. NRTIs act by inhibiting the HIV reverse transcriptase enzyme, thereby blocking viral DNA synthesis. Used together, these agents act synergistically to suppress viral replication and delay the progression of HIV infection. Clinical trials have shown this combination is more effective in treatment-naive patients than NRTIs alone or amprenavir monotherapy[1][2][5][7]. The regimen is generally well tolerated, with gastrointestinal disturbances and skin rash being the most common adverse events. Amprenavir can be co-administered with other antiretroviral agents for enhanced efficacy, but its use in combination requires careful consideration of drug-drug interactions, particularly with agents metabolized by CYP3A4[5][7].
