**Amrubicin + trastuzumab** is an investigational combination therapy comprising amrubicin, a synthetic 9-aminoanthracycline small molecule chemotherapeutic agent, and trastuzumab, a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2). Amrubicin acts primarily as a DNA topoisomerase II inhibitor, inducing cytotoxicity in rapidly dividing tumor cells. Trastuzumab binds with high specificity to HER2 receptors on cancer cells, blocking HER2 signaling, promoting antibody-dependent cell-mediated cytotoxicity (ADCC), and thereby inhibiting tumor cell proliferation. Preclinical studies suggest this combination has synergistic antitumor effects, particularly in HER2-positive cancers, by leveraging both a cytotoxic mechanism and targeted immunologic inhibition[1][2].
