AMT-116 is a first-in-class antibody-drug conjugate (ADC) developed by Multitude Therapeutics Inc. It comprises a humanized anti-CD44v9 IgG1 antibody conjugated to KL610023, a novel belotecan-derived topoisomerase I inhibitor, via a cleavable hydrolysable linker. The drug targets CD44v9, which is highly expressed in various solid tumors, and delivers the topoisomerase I inhibitor payload to exert its antitumor activity. It is currently being evaluated in Phase 1/2 clinical trials for advanced solid tumors.
