Drug intelligence / Profile preview
AMT-116 + ivonescimab is an investigational combination therapy being evaluated for the treatment of advanced non-small cell lung cancer (NSCLC). The combination consists of AMT-116, a first-in-class antibody-drug conjugate (ADC) targeting CD44v9, and ivonescimab (AK112), a first-in-class bispecific antibody targeting programmed cell death protein 1 (PD-1) and vascular endothelial growth factor A (VEGF-A). AMT-116 comprises a humanized anti-CD44v9 IgG1 antibody conjugated to KL610023, a belotecan-derivative topoisomerase I inhibitor payload, via a hydrolysable linker. Ivonescimab is a humanized IgG1 bispecific antibody that simultaneously blocks PD-1 and VEGF-A pathways. By combining these agents, the regimen aims to deliver targeted cytotoxic therapy to CD44v9-expressing tumor cells while simultaneously enhancing anti-tumor immune responses and inhibiting angiogenesis. The combination is being evaluated in a Phase Ib/II clinical trial (NCT07590531) sponsored by Multitude Therapeutics.
