AMT-253 is a first-in-class antibody-drug conjugate (ADC) designed to target MUC18 (also known as Melanoma Cell Adhesion Molecule, MCAM, or CD146), a cell surface glycoprotein overexpressed in various solid tumors. Developed by Multitude Therapeutics, the drug consists of a humanized IgG1 antibody with high affinity for MUC18 conjugated to exatecan, a potent topoisomerase I (TOP1) inhibitor. The conjugation utilizes a proprietary protease-cleavable self-immolative T moiety linker with a drug-to-antibody ratio (DAR) of 8. AMT-253 is currently being evaluated in Phase 1/2 clinical trials for the treatment of advanced solid tumors, specifically focusing on melanoma and gynecologic cancers such as ovarian, cervical, and endometrial cancers. It has demonstrated promising antitumor activity and a manageable safety profile in early-stage clinical settings.
