AMT-253 CAR-T cells are an autologous chimeric antigen receptor (CAR) T-cell therapy designed to target cells expressing the melanoma-associated antigen MUC18 (also known as CD146 or MCAM). Developed for the treatment of advanced solid tumors, particularly melanoma, this therapy involves genetically modifying a patient's own T-lymphocytes to express a receptor that recognizes MUC18, a glycoprotein highly expressed in several subtypes of melanoma and its associated tumor vasculature. Upon infusion, the engineered T cells bind to MUC18-positive tumor cells, initiating a targeted immune response and direct cytotoxic cell death. Clinical investigation for this modality has explored both intravenous and intratumoral routes of administration, specifically within dose-escalation trials conducted by Beijing Immunochina Medical Science & Technology.
