AMT-862 is an investigational antibody-drug conjugate (ADC) developed by Multitude Therapeutics for the treatment of HER3-expressing solid tumors. It consists of a monoclonal antibody targeting Human Epidermal Growth Factor Receptor 3 (HER3/ErbB3) conjugated to a potent topoisomerase I (TOP1) inhibitor payload, specifically exatecan or a derivative. HER3 is frequently overexpressed in various cancers, including lung, breast, and colorectal cancers, and is associated with poor prognosis and resistance to other therapies. Upon binding to HER3 on the tumor cell surface, the ADC is internalized, and the TOP1 inhibitor is released, leading to DNA damage and apoptosis. AMT-862 is currently in the IND-enabling stage of development.
