AMTX-100 is a first-in-class, cell-penetrating synthetic peptide under development by Amytrx Therapeutics for the treatment of inflammatory and autoimmune diseases, with its lead indication being atopic dermatitis. The drug is a 28-amino acid chimeric polypeptide bioengineered from sequences of human fibroblast growth factor 4 (FGF4) and nuclear localization sequences from transcription factors such as NF-kappaB. Its mechanism involves modulating the nuclear transport of stress-responsive transcription factors (SRTFs) and metabolic transcription factors by competing for binding to importin alpha5/beta1 complexes, thereby attenuating inflammatory gene expression without broadly suppressing essential cellular functions. This targeted modulation aims to reduce inflammation while minimizing side effects commonly seen with other anti-inflammatory agents. Clinical development has reached Phase I/II for atopic dermatitis, with additional studies in acne, psoriasis, rosacea, herpes zoster, cutaneous lupus erythematosus (Phase I), and preclinical research in a wide range of other inflammatory conditions including rheumatoid arthritis and non-alcoholic steatohepatitis[1][2][6][7][8].
