Overview

Amuc_1434 is a recombinant aspartic protease protein derived from the gut bacterium Akkermansia muciniphila. It has been studied primarily for its antitumor effects in colorectal cancer models. Mechanistically, Amuc_1434 degrades Mucin2 (MUC2), a major component of the intestinal mucus layer that is highly expressed in certain colorectal cancers. This degradation leads to inhibition of tumor cell proliferation, induction of cell cycle arrest at G0/G1 phase, upregulation of p53 expression, increased mitochondrial reactive oxygen species (ROS), and loss of mitochondrial membrane potential. Critically, it promotes apoptosis via activation of both death receptor and mitochondrial pathways by upregulating tumor-necrosis-factor-related apoptosis-inducing ligand (TRAIL). Additionally, studies indicate that outer membrane vesicles containing Amuc_1434 can modulate PD-L1 expression and enhance CD8+ T cell-mediated antitumor immunity[1][2][3][7].
