AMXT-1501 is a novel, orally bioavailable small molecule polyamine transport inhibitor developed for cancer therapy. It acts by blocking the uptake of polyamines from the bloodstream into tumor cells and the tumor microenvironment, thereby reducing intracellular polyamine concentrations. This mechanism inhibits tumor cell proliferation, induces apoptosis, and may reverse polyamine-mediated immune suppression in the tumor microenvironment. By depleting polyamines—molecules essential for cell growth and division—AMXT-1501 can both directly suppress cancer cell growth and enhance anti-tumor immune responses. The drug is being investigated primarily in combination with difluoromethylornithine (DFMO), a polyamine synthesis inhibitor, as an orthogonal immunometabolic approach to treat various solid tumors and pediatric cancers[3][5][6][8].
