AMY-106 is a fourth-generation compstatin-based complement C3 inhibitor being developed by Amyndas Pharmaceuticals for the treatment of age-related macular degeneration (AMD), geographic atrophy (GA), and intermediate AMD. It is a synthetic cyclic peptide designed for local ocular administration via intravitreal injection to treat complement-mediated retinal diseases. AMY-106 features enhanced pharmacokinetic properties and improved solubility compared to earlier generation compstatins, with preclinical studies in non-human primates demonstrating an extended residence time in the vitreous of over three months following a single injection. The drug works by binding to and inhibiting complement component C3, thereby blocking complement activation in the eye and potentially preventing or reducing the rate of retinal cell death and the progression of intermediate AMD to geographic atrophy or wet AMD.
