Amycretin is a unimolecular, long-acting dual agonist of the glucagon-like peptide 1 (GLP-1) receptor and the amylin receptor, under development by Novo Nordisk for the treatment of obesity, overweight, and type 2 diabetes. Unlike some existing GLP-1 agonists that require injection, amycretin is being developed in both oral and subcutaneous formulations. Its mechanism involves mimicking two key hormones—GLP-1 to enhance insulin secretion and promote satiety, and amylin to regulate appetite and slow gastric emptying—resulting in reduced food intake and improved glucose control. Clinical trials have shown significant weight loss effects with up to 22% reduction in body weight over 36 weeks at higher doses. The drug consists of two covalently linked peptides (analogs of GLP-1 and amylin), with oral formulations using salcaprozate sodium as a permeation enhancer[1][5][6].
