AmyTrap-3 is a patented, synthetic peptide-based therapeutic developed for the treatment of Alzheimer's disease. It consists of polymer-bound short peptides, specifically a retro-inverso (RI) peptide designed to bind and sequester amyloid-beta (Aβ) peptides. The molecule is conjugated to albumin via a polymer moiety, enhancing its stability and circulatory half-life. Its mechanism involves binding to the GXXXG motif in Aβ, which is critical for Aβ misfolding and aggregation. By capturing circulating Aβ in the periphery rather than entering the brain directly, AmyTrap-3 acts as a "peripheral sink," promoting efflux of Aβ from the brain into circulation and thereby reducing amyloid plaque burden in preclinical Alzheimer's disease models. This approach has shown efficacy in reducing brain plaques and improving cognitive function in animal studies without inducing immunogenicity or significant toxicity[2][4][5][6]. The drug remains at the preclinical development stage[1][7].
