AN-1792 is an investigational active immunotherapy developed for the treatment of Alzheimer's disease. It consists of a synthetic full-length beta-amyloid (Aβ) 1–42 peptide formulated with the QS-21 adjuvant. The therapeutic rationale was to induce an immune response that would promote clearance of amyloid plaques from the brain, a hallmark pathology in Alzheimer's disease. Preclinical studies demonstrated that immunization with Aβ1–42 could prevent or reverse neuropathological features associated with Alzheimer's disease in animal models. In clinical trials, AN-1792 showed some evidence of reducing amyloid plaque burden and decreasing cerebrospinal fluid tau levels in antibody responders; however, development was halted during Phase 2a due to cases of meningoencephalitis observed in approximately 6% of treated patients[1][2][3][5]. The drug was never approved and its development was terminated.
