Overview

AN-215 is a cytotoxic drug initially developed by Æterna Zentaris, Inc. It functions through a multi-targeted mechanism, acting as a DNA intercalator, a gastrin-releasing peptide receptor (GRPR) antagonist, and a topoisomerase II inhibitor. This approach is designed to disrupt DNA replication and transcription, block growth-promoting signals mediated by GRPR, and inhibit DNA unwinding, thereby suppressing cancer cell proliferation. The drug is a cytotoxic analog of bombesin/GRP, containing 2-pyrrolino-Dox. It was investigated for its potential in treating neoplasms, specifically in women with ovarian or endometrial cancer, where it demonstrated disease stabilization and objective responses in Phase I clinical trials. Its development has since been discontinued.
