Drug intelligence / Profile preview
AN-5162 is an orally bioavailable, AI-designed small-molecule inhibitor developed by Anew Therapeutics, a spinout of ByteDance's AI drug-discovery unit Anew Labs. It is designed to target and potently block the full spectrum of Interleukin-17 (IL-17) dimers, including IL-17AA, IL-17AF, and IL-17FF. Unlike traditional small-molecule IL-17 inhibitors that are restricted to IL-17A, AN-5162 achieves pan-inhibition of all three dimers, offering biologics-like efficacy with the convenience of oral dosing. In preclinical studies, AN-5162 demonstrated high-affinity binding and robust functional antagonism, with Ki values of <0.1 nM for IL-17A and 6.2 nM for IL-17FF, and showed dose-dependent suppression of IL-17-driven cytokine production in vivo. It is currently in preclinical development for the treatment of autoimmune and inflammatory diseases, such as psoriasis and ankylosing spondylitis.
