AN0025 + pembrolizumab is a combination investigational therapy studied in advanced solid tumors, including non-small cell lung cancer, for patients who have progressed on prior immunotherapy. AN0025 (formerly E7046) is a *selective oral antagonist of the prostaglandin E2 receptor subtype 4 (EP4)*, designed to block prostaglandin E2-mediated immunosuppression within the tumor microenvironment and potentiate anticancer immune responses. Pembrolizumab is a *monoclonal antibody checkpoint inhibitor* targeting the *programmed cell death protein 1 (PD-1) receptor*, and is established as an immunotherapy to reactivate anti-tumor T cell responses. The combination aims to overcome immunotherapy resistance by simultaneously blocking prostaglandin E2 signaling and PD-1 inhibitory signaling. AN0025 was licensed from Eisai and is being developed by Adlai Nortye, with clinical collaboration with Merck for pembrolizumab[2][5][7].
