Overview

AN0128 is a boron-containing small molecule, specifically a borinic acid picolinate ester, with both antibacterial and anti-inflammatory properties. It was developed by Anacor Pharmaceuticals as a topical therapeutic candidate for dermatological conditions such as atopic dermatitis, acne vulgaris, psoriasis, and rosacea. The compound demonstrates broad-spectrum activity against Gram-positive bacteria (notably Propionibacterium acnes), making it relevant for skin infections and inflammatory skin diseases. Mechanistically, AN0128 inhibits the release of pro-inflammatory cytokines including tumor necrosis factor alpha (TNF-alpha), without suppressing normal immune responses[1][2][4][5][6]. Clinical studies have shown promising results in moderate atopic dermatitis[5].
