Overview

AN2-502998 (formerly known as AN15368) is an orally bioavailable, boron-containing heterocyclic small molecule under development for the treatment of chronic Chagas disease. It acts as a selective inhibitor of the cleavage and polyadenylation specificity factor subunit 3 (CPSF3) protein in *Trypanosoma cruzi*, the parasite responsible for Chagas disease. This mechanism is similar to that of acoziborole, which has shown high cure rates in related trypanosome infections. Preclinical studies have demonstrated that AN2-502998 can achieve curative activity in non-human primates with long-term, naturally acquired chronic *T. cruzi* infection—a model closely resembling human disease progression. There are currently no FDA-approved therapies for chronic Chagas disease in adults; existing treatments are limited by efficacy and tolerability issues[1][3][6].
