AN2728 ointment vehicle refers to the inactive topical formulation used as a comparative control (placebo) in clinical trials evaluating AN2728 (crisaborole). AN2728 is a boron-based small molecule phosphodiesterase-4 (PDE4) inhibitor developed by Anacor Pharmaceuticals (later acquired by Pfizer). While the active drug, crisaborole (brand name Eucrisa), is primarily indicated for atopic dermatitis, it was also investigated in Phase 1 and Phase 2 trials for the treatment of plaque psoriasis. The vehicle ointment contains the same excipients as the active 2% or 5% AN2728 ointment but lacks the active PDE4 inhibitor, serving to isolate the therapeutic effect of the drug from the emollient properties of the ointment base in clinical efficacy and safety assessments.
