Overview

AN2898 is a boron-containing small molecule developed as a topical anti-inflammatory agent for the treatment of atopic dermatitis and psoriasis. It acts as a competitive and reversible inhibitor of phosphodiesterase type 4 (PDE4), with additional inhibitory activity against PDE7. By inhibiting these enzymes, AN2898 reduces the production of pro-inflammatory cytokines such as tumor necrosis factor alpha (TNFα), interleukin (IL)-12, and IL‑23. Clinical studies demonstrated that topical application of AN2898 led to significant improvements in lesion severity scores in patients with mild-to-moderate atopic dermatitis compared to vehicle controls. The drug was originally developed by Anacor Pharmaceuticals and later by Pfizer following acquisition. Despite promising early-phase results showing efficacy and safety in Phase 2 trials for atopic dermatitis and psoriasis, development appears to have been discontinued[5][6][7][9].
