Overview

AN4005 is an orally available, small-molecule inhibitor of programmed cell death ligand 1 (PD-L1), designed to disrupt the interaction between PD-L1 and programmed cell death protein 1 (PD-1). By blocking this immune checkpoint pathway, AN4005 aims to restore antitumor immune responses. Unlike monoclonal antibody-based PD-(L)1 inhibitors, AN4005 offers potential advantages such as oral administration, lower production costs, improved tumor penetration, and reduced immunogenicity. Preclinical studies have shown that AN4005 is well tolerated and exhibits tumor growth inhibition comparable to FDA-approved anti-PD-L1 antibodies in an immune-dependent manner. The drug is being developed primarily for advanced solid tumors and is currently undergoing Phase I clinical trials to determine its recommended Phase 2 dose and maximum tolerated dose[1][2][4][6][8].
