AN8025 is a first-in-class, tri-specific antibody fusion protein designed for cancer immunotherapy. It is derived from a clinically validated and approved anti-PD-L1 antibody, with functionally optimized CD86 (CD86v) and LAG3 (LAG3v) variants fused to the C- and N-terminus of the heavy chain, respectively. The drug acts as both a T cell and antigen-presenting cell (APC) modulator. Mechanistically, the LAG3 component stimulates differentiation and maturation of APCs; CD86 and PD-L1 components synergistically amplify tumor-specific T cell responses. This multifaceted approach enhances immune function by improving both quantity/quality of APCs and inducing robust PD-L1-dependent T cell activation—without causing unconditional activation in human primary T cells where PD-L1 levels are low. Preclinical studies have shown potent antitumor efficacy in syngeneic and humanized mouse models[2][4][6][8].
