ANA-12 is a selective, small-molecule, non-competitive antagonist of the tropomyosin receptor kinase B (TrkB), which is the primary receptor for brain-derived neurotrophic factor (BDNF). Discovered in 2011 by researchers at Université Paris and Inserm, the compound binds to TrkB at both high- and low-affinity sites, effectively blocking BDNF-induced TrkB activation without interfering with TrkA or TrkC signaling. ANA-12 is capable of crossing the blood-brain barrier and has demonstrated rapid antidepressant and anxiolytic-like effects in various animal models of mood disorders. While it is widely utilized as a pharmacological research tool to investigate the BDNF/TrkB pathway in conditions such as depression, anxiety, and Alzheimer's disease, it is not currently an approved therapeutic agent for human use.
