ANA-targeted dactinomycin is an experimental nanoparticle formulation being developed by NanoSmart Pharmaceuticals for the treatment of Ewing's sarcoma. The drug consists of the cytotoxic chemotherapy agent dactinomycin (actinomycin D) encapsulated within a proprietary nanoemulsion or liposomal vehicle. This delivery system is conjugated with a human-derived anti-nuclear antibody (ANA) that specifically targets nuclear antigens. These antigens are typically sequestered within healthy cells but become accessible in the necrotic regions of solid tumors. By targeting these areas of necrosis, the ANA-targeted formulation is designed to accumulate dactinomycin preferentially at the tumor site, potentially reducing systemic toxicity and allowing for the administration of higher therapeutic doses. The FDA granted this formulation Orphan Drug Designation for Ewing's sarcoma in July 2015 based on its potential for clinical superiority over conventional dactinomycin.
