Anagin is developing a first-in-class small molecule modulator of the N-methyl-D-aspartate receptor (NMDAR) signaling complex, specifically designed to treat Post-Traumatic Stress Disorder (PTSD) and related psychiatric conditions. The therapeutic strategy involves disrupting the protein-protein interaction between the NMDAR, the scaffolding protein PSD-95, and neuronal nitric oxide synthase (nNOS). By uncoupling this signaling complex, the drug reduces excessive nitric oxide production in specific brain regions associated with fear and injury, without directly blocking the NMDAR ion channel. This targeted approach aims to correct the brain's ongoing fear response while avoiding the learning deficits, hallucinations, and ataxia typically associated with traditional NMDAR antagonists. The program is being explored for multiple indications, including depression, substance use disorder, chronic pain, obsessive-compulsive disorder, and various anxiety disorders.
