Overview

Analog D is a novel synthetic ceramide analog developed as a potential therapeutic agent for chemotherapy-resistant and hormone therapy-resistant breast cancers. Derived from the lead compound N-octanoyl-D-erythro-sphingosine (C8-ceramide), Analog D features either an enol or amide backbone substitution designed to mimic endogenous long-chain ceramides and induce apoptosis. In preclinical evaluations, Analog D demonstrated potent antiproliferative activity, particularly in the triple-negative breast cancer cell line MDA-MB-231, where it exhibited a low IC50 of 460 nM in clonogenic proliferation assays. This represents a significant increase in potency compared to standard C8-ceramide, highlighting its potential as a targeted therapy to overcome endocrine and chemotherapy resistance in breast cancer.
