ANANAS-ApoE is an experimental nanoparticle-based drug delivery platform designed to facilitate the transport of therapeutic agents across the blood-brain barrier (BBB). The platform, known as ANANAS (avidin-nucleic-acid-nanoassembly), consists of a core assembly of avidin and nucleic acids, which is functionalized with apolipoprotein E (ApoE) to potentially target receptors such as LRP1 or LDLR on brain endothelial cells. Research in murine models of amyotrophic lateral sclerosis (ALS) has shown that while the base ANANAS assembly can transiently cross the BBB, the addition of ApoE does not significantly enhance this penetration in the tested SOD1G93A model. The platform has been evaluated for the delivery of cargo such as dexamethasone, though effective release of the therapeutic payload within the central nervous system remains a challenge.
