**ANANAS-Dex** is an experimental dexamethasone nanoconjugate in which dexamethasone is covalently tethered to an avidin-nucleic-acid nanoassembly carrier through an acid-reversible hydrazone linker. The formulation is designed to concentrate the glucocorticoid in selected tissues and release free dexamethasone after uptake into acidic intracellular compartments. In murine studies, it demonstrated liver tropism and preclinical activity in autoimmune hepatitis models; it has also been evaluated in pulmonary fibrosis, amyotrophic lateral sclerosis, and cholangitis-related animal studies. In the SOD1G93A amyotrophic lateral sclerosis mouse model, the carrier transiently crossed the blood-brain barrier but dexamethasone concentrations in central nervous system tissues remained below detection limits, indicating inadequate payload release in the CNS.
