Overview

Anaplastic lymphoma kinase-directed chimeric antigen receptor t-cell therapy is an investigational cell therapy being developed by ALKemist Bio for the treatment of neuroblastoma. The therapy consists of T cells (either autologous or allogeneic) that are genetically engineered ex vivo to express a chimeric antigen receptor (CAR) targeting the extracellular domain of anaplastic lymphoma kinase (ALK). ALK is a receptor tyrosine kinase that is highly expressed as a membrane-bound protein on the surface of most neuroblastoma cells but is largely absent in normal adult tissues, making it a suitable target for immunotherapy. These CAR-T cells are designed to recognize membrane-expressed ALK and mediate T cell-driven cytotoxicity against tumor cells. Currently in preclinical development, this therapy is part of a dual approach by ALKemist Bio to target ALK-positive cancers, complementing their TCR-T platform which targets intracellular ALK fusion proteins.
