Overview

AnAPN1 is a recombinant protein vaccine candidate designed as a pan-malaria transmission-blocking vaccine. It is based on the highly conserved luminal midgut surface glycoprotein alanyl aminopeptidase N (AnAPN1) from the mosquito species *Anopheles gambiae*. The vaccine works by eliciting antibodies in humans that target the AnAPN1 protein in mosquitoes. When a mosquito feeds on vaccinated blood, these antibodies block the development of *Plasmodium* parasites within the mosquito gut, thereby preventing further transmission of malaria to other individuals. Unlike traditional malaria vaccines that target parasite antigens directly in humans, AnAPN1 represents a "mosquito-based" approach to interrupting disease spread[2][4]. The recombinant protein is expressed in *Escherichia coli* and formulated with adjuvants such as Alhydrogel for enhanced immunogenicity[3]. Clinical development has included preclinical studies and first-in-human trials[2][4].
