Overview

Anastrozole + goserelin is a combination therapy used primarily in premenopausal women with hormone receptor-positive, recurrent, or advanced breast cancer. Anastrozole is a selective aromatase inhibitor that blocks the conversion of androgens to estrogens by inhibiting the aromatase enzyme, leading to reduced estrogen levels. Goserelin is a synthetic analog of luteinizing hormone-releasing hormone (LHRH) that acts as an agonist at the gonadotropin-releasing hormone receptor; chronic administration results in downregulation of pituitary gonadotropin secretion and suppression of ovarian estradiol production. The combination achieves profound estrogen deprivation by both suppressing ovarian function (goserelin) and blocking peripheral estrogen synthesis (anastrozole). This regimen is considered when tamoxifen is ineffective or contraindicated[1][2][4][5][8].
