Anastrozole + triptorelin is a combination therapy consisting of anastrozole, a nonsteroidal aromatase inhibitor, and triptorelin, a synthetic decapeptide analog of gonadotropin-releasing hormone (GnRH agonist). This combination is primarily used in the treatment of endocrine-responsive early-stage breast cancer in premenopausal women at high risk of recurrence and has also been studied for use in pediatric endocrinology to manage early puberty and improve height outcomes. Anastrozole works by inhibiting the aromatase enzyme, thereby reducing estrogen synthesis from androgen precursors. Triptorelin suppresses ovarian function by downregulating pituitary GnRH receptors, leading to decreased secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), which results in reduced ovarian estrogen production. The combined effect achieves profound estrogen suppression suitable for conditions where estrogen drives disease progression[2][3][5][7].
