ANAVEX1-41 is a novel small molecule drug candidate developed by Anavex Life Sciences for the treatment of neurodegenerative disorders, with a primary focus on Alzheimer's disease. It acts as an agonist at sigma-1 receptors and also modulates muscarinic receptors, sodium channels, and chloride channels. Preclinical studies have demonstrated that ANAVEX1-41 possesses potent neuroprotective and anti-amnesic properties at extremely low doses, reversing memory and learning deficits in animal models of Alzheimer's disease induced by amyloid-beta peptide. The compound prevents oxidative stress, neuronal apoptosis (by blocking caspase-3 expression), and loss of hippocampal neurons. Its mechanism involves synergistic activation of sigma-1 and muscarinic receptors to regulate calcium mobilization in neuronal cells, thereby protecting against endoplasmic reticulum stress-induced cell death[3][4][5][6][7][10].
